Pharmacological Actions of Intraventricular Clonidine and Methoxamine on Heart Rate of Rabbits

Abstract
|
|
|
1) Effects of intraventricular clonidine and methoxamine, a-adrenoceptor agonists, on heart rate of rabbits were investigated.
2) Clonidine produced dose-dependent decrease of heart rate, which was competitively inhibited by intraventricular yohimbine and phentolamine.
3) Methoxamine produced dose-dependent decrease of heart, which was not affected by intraventricular yohimbine but competitively inhibited by intraventricular phentolamine. (The dose to inhibit the methoxamine effect was greater than that to inhibit the clonidine one.)
4) Intraventricular phenoxybenzamine produced persistent inhibition of the effects of clonidine and methoxamine.
5) Following pretreatment with intraventricular phentolamine,.-phenoxybenzamine was unable to inhibit the methoxamine effect.
6) It is inferred that clonidine produced the bradycardiac effect by acting on a2-adrenoceptors in the brain and methoxamine on al-adrenoceptors.
